Adventitious agent contamination risk mitigation: engineering MMV virus resistance into CHO cells

چکیده
Background The introduction of animal origin free (AOF) media has significantly reduced the incidence of adventitious virus contamination in biological production systems. Nevertheless, contamination by the parvovirus Mouse Minute Virus (MMV) remains a continuing challenge. Although infrequent, infection of a fermenter can be catastrophic for a manufacturer, and can also have a potential impact on drug supply, patient safety and have regulatory implications. In this work, we evaluated engineering Chinese Hamster Ovary (CHO) cell lines to create a new host cell line that

Adventitious agents and live viral vectored vaccines: Considerations for archiving samples of biological materials for retrospective analysis.
Vaccines are one of the most effective public health medicinal products with an excellent safety record. As vaccines are produced using biological materials, there is a need to safeguard against potential contamination with adventitious agents. Adventitious agents could be inadvertently introduced into a vaccine through starting materials used for production. Therefore, extensive testing has be...

Viral safety testing is a critical part of the Chemistry, Manufacturing and Control (CMC) of biologics throughout the development and release of commercial products. A major safety risk to humans and a financial burden for biopharmaceutical companies, viral contamination can arise from the contamination of cell substrates or the adventitious introduction of a virus during production. Covance an...

Emerging Infectious Diseases Vol. 7, No. 3 Supplement, June 2001 562 A discussion of adventitious agents and vaccines is pertinent in the context of emerging infectious diseases. Many novel vaccines are produced in animal cell substrates, and emerging infectious diseases may theoretically be transmitted from animals to humans through these vaccines. The challenge of identifying potential advent...

Background and Aims: Adventitious agents, especially viral agents are among the most important concerns in viral vaccines. Viral contamination of biological products may arise from the original source of the reagents such as, serum, trypsin, animal or human derived media components or cell culture and working seed or cross contamination of vaccine during production. In this study, adventitious ...

ABSTRACT Five tropical maize lines were tested and compared with the susceptible control line B73 for resistance to Maize stripe virus (MStV) and Maize mosaic virus (MMV), both propagatively transmitted by the planthopper Peregrinus maidis (Homoptera: Delphacidae). Resistance to each virus was evaluated separately by artificial inoculations with planthoppers viruliferous for either one virus or...
